OBJECTIVE@#To investigate the regulatory effect of iridoid glycoside of radix scrophulariae (IGRS) on endoplasmic reticulum stress induced by oxygen-glucose deprivation and reperfusion @*METHODS@#Rat pheochromocytoma PC12 cells were pretreated with IGRS (50, 100, 200 μg/mL) for 24h, and the @*RESULTS@#The damage caused by OGD/R to PC12 cells was significantly reduced by IGRS, with significant effect on increasing survival rate and reducing LDH release (all @*CONCLUSIONS@#IGRS has neuroprotective effect, which may alleviate cerebral ischemia-reperfusion injury by regulating SERCA2, maintaining calcium balance, and inhibiting endoplasmic reticulum stress-mediated apoptosis.

ABSTRACT The objective of this study was to identify thyroid hormones and to examine their putative site of synthesis in Achatina fulica snails. For this purpose, radioimmunoassays were performed for T3 and T4 before and after long starvation with or without hemolymph deproteinization. Sodium/iodide symporter activity in vivo was analyzed through 125I administration with and without KClO4 pretreatment. Only T4 was detected, and its concentration decreased due to starvation or deproteinization. However, high-performance liquid chromatography analysis also showed the presence of T2 and T3 apart from T4, but rT3 was not detected in the A. fulica hemolymph. The sodium/iodide symporter activity was greater in cerebral ganglia than digestive gland, but KClO4 treatment did not inhibit iodide uptake in any of the tissues analyzed. Altogether, our data confirm for the first time the presence of thyroid hormones in A. fulica snails and suggest their participation in the metabolism control in this species, although the putative site of hormone biosynthesis remains to be elucidated.

This study evaluated the effect of oral glucosamine and intramuscular injection (IM) of snail mucin on the progression of experimental osteoarthritis (OA) in dogs. Twenty adult mongrels with mean body weight (12.4±1.8 kg) were used. Experimental OA was induced surgically using the groove model. The dogs were randomly divided into three groups following radiographic evidence of OA. Group one (control) comprised of ten dogs treated with normal saline twice weekly for four weeks following OA. Group two comprised of five dogs treated with 10mg/kg of oral glucosamine daily for four weeks. Group three comprised of five dogs treated with 5mg/kg intramuscular injection of 5% solution of snail mucin twice weekly for four weeks. Blood was obtained from the cephalic vein before surgical arthrotomy, after surgical arthrotomy, immediately after radiographic confirmation of OA (Week 0) and at two weeks interval up to 4 weeks of treatment. Efficacy of the drugs was assessed by changes in plasma IL-6 and MMP-3, while safety was determined using the changes in packed cell volume (PCV), total white blood cell counts (WBC) and observable adverse reactions associated with the administration of the drugs. In this study, the PCV and WBC did not differ significantly (P> 0.05) from the control group. Plasma IL-6 and MMP-3 were significantly (P< 0.05) lower both in glucosamine-treated and snail mucin-treated dogs up to week 4 of treatment when compared with the control group. However, there were no significant (P > 0.05) differences in IL-6 and MMP-3 between the two treatment groups. In addition, painful swelling at the site of injection was observed in dogs treated with snail mucin, while no adverse reaction was observed in dogs treated with oral glucosamine. It was therefore concluded that both oral glucosamine and IM injection of snail mucin comparably modified the progression of OA. However, owing to the adverse reaction noted with IM injection of snail mucin, further study is required to determine the most appropriate route of administration.


Se evaluaron los efectos de la glucosamina oral y la inyección intramuscular (IM) de mucina de caracol en la progresión de la osteoartritis (OA) experimental en perros. Fueron utilizados 20 perros mestizos adultos con un peso medio de 12,4±1,8 kg. La OA experimental se indujo quirúrgicamente mediante el modelo de ranura. Los animales se dividieron aleatoriamente en tres grupos después de la evidencia radiográfica de OA. El grupo 1 (control, 10 perros) fue tratado con una solución salina normal dos veces por semana durante cuatro semanas. El grupo 2 (5 perros) fue tratado con 10 mg/kg de glucosamina oral al día por cuatro semanas, y el grupo 3 (5 perros) fue tratado con 5 mg/kg IM de una solución de mucina de caracol al 5% dos veces por semana durante cuatro semanas. Se obtuvieron muestras de sangre desde la vena cefálica previo a la artrotomía quirúrgica, después de la artrotomía e inmediatamente después de la confirmación radiográfica de OA (semana 0), y en el intervalo de dos semanas hasta cuatro semanas de tratamiento. La eficacia de los fármacos se evaluó por los cambios plasmáticos de IL-6 y MMP-3, mientras que la seguridad, se determinó por los cambios en el volumen del hematocrito (VH), el recuento total de glóbulos blancos (RGB), y la observación de reacciones adversas asociadas a la administración de fármacos. El VH y RGB no difirieron significativamente (P>0,05) en el grupo control. Los niveles de IL-6 y MMP-3 plasmática fueron significativamente más bajas (P<0,05) en los perros tratados con glucosamina y mucina de caracol hasta 4 semanas, en comparación con el grupo control. Sin embargo, no hubo diferencias significativas (P>0,05) en la IL-6 y MMP-3 entre los dos grupos de tratamiento. Además, se observó un edema doloroso en el sitio de inyección de los perros tratados con mucina de caracol. En los perros tratados con glucosamina oral no se observó reacción adversa. Se concluye que tanto la glucosamina oral y la inyección IM de mucina de caracol modifican comparablemente la progresión de OA. Sin embargo, debido a la reacción adversa observada con la inyección IM de mucina caracol, se necesitan estudios adicionales para determinar la vía de administración más adecuada.

Achatina fulica's secretion has been recently related to antibacterial, antifungal, and cicatricial properties, and it is influenced by the kind of food offered. Therefore, this study investigated the healing potential of dressing films based on mucous secretion of Achatina fulica. Thus, surgical wounds performed in black wistar rats were dressed with films based on collagen, and on mucous secretion of A. fulica fed with Lactuca sativa; undresses worked as wounds control. After 3, 7, 14 and 21 days the animals were euthanized, and the wounds were microscopically evaluated. On the 3rd and 7th days dressing films based on mucus provided acceleration of the formation maturation of granulation tissue, better epithelization rates, and more rapid replacement of type III for type I collagen fibers. On the 14th and 21st days, these dressings induced more intense deposition and better architectural disposition of type I collagen fibers, and hastened the regeneration of cutaneous phaneros. Dressings obtained from A. fulica fed with Lactuca sativa provided the most expressive results. This study suggests that films produced with mucous secretion of A. fulica can be successfully employed as wound dressing, particularly if snails are fed with Lactuca sativa.


A la secreción mucoproteica del Achatina fulica se le han asignado propiedades antibacterianas, antifúngicas y cicatriciales que pueden estar influidas por el tipo de alimento que se ofrece a este especimen. Este estudio investigó el potencial curativo de películas basadas en la secreción mucosa de Achatina fulica alimentaba con diferentes tipos de plantas. Fueron tratadas heridas provocadas en el dorso de ratas Wistar con películas de colágeno y películas realizadas a partir de la secreción mucosa de A. fulica, alimentados con plantas de lechuga (L. sativa), utilizando como parámetro de comparación la curación del grupo de control. Después de 3, 7, 14 y 21 días los animales fueron sacrificados y las heridas fueron evaluadas microscópicamente. En los días 3 y 7, las heridas tratadas con moco mostraron mejores tasas de formación y maduración del tejido de granulación, epitelización, y más rápido recambio de colágeno tipos III y I. A los días 14 y 21, hubo una intensa deposición del colágeno tipo I y aceleración en la regeneración de la piel. Los resultados obtenidos a partir de A. fulica alimentados con plantas de lechuga (L. sativa) mostraron mejores resultados. Este estudio sugiere que las películas producidas con secreción mucosa de A. fulica pueden ser utilizadas con éxito como apósito, especialmente si los caracoles se alimentan con plantas de lechuga L. sativa.

A 70-yr-old woman visited our hospital for shortness of breath. Chest CT showed ground glass opacity and traction bronchiectasis at right middle, lower lobe and left lingular division. Video-assisted thoracic surgical biopsy at right lower lobe and pathologic examination revealed mixed dust pneumoconiosis. Polarized optical microscopy showed lung lesions were consisted of silica and carbon materials. She was a housewife and never been exposed to silica dusts occupationally. She has taken freshwater snails as a health-promoting food for 40 yr and ground shell powder was piled up on her backyard where she spent day-time. Energy dispersive X-ray spectroscopy of snail shell and scanning electron microscopy with energy dispersive x-ray spectroscopy of lung lesion revealed that silica occupies important portion. Herein, we report the first known case of silicosis due to chronic inhalation of shell powder of freshwater snail.

Fatty acids composition of the marine snails Phyllonotus pomum and Chicoreus brevifrons (Muricidae). Muricid species of P. pomum and C. brevifrons are of economic importance in the Caribbean. This study includes a comparative evaluation of fatty acid content in the total lipid composition of Phyllonotus pomum and Chicoreus brevifrons. Snail samples were collected during the rainy, dry and transition seasons, in Punta Arena, Sucre (Venezuela). Total lipids were extracted and the specific fatty acid contents were analyzed by gas chromatography. Lipid concentrations varied between 0.87 and 1.85%, with minimum and maximum values corresponding to C. brevifrons collected during rainy and dry seasons, respectively. In the case of total lipids, a high concentration of unsaturated fatty acids (57.21-70.05%) was observed followed by saturated fatty acids (20.33- 31.94%), during all seasons. The polyunsaturated occurred in higher proportion among the unsaturated fatty acids, except for P. pomum which showed higher proportion of monounsaturated fatty acids (38.95%) during the transition season. The prevailing fatty acids were: C14:0, C16:0, C18:0, C20:1, C22:1 ω-11, C22:1 ω-9, C18:3 ω-3, C20:5 ω-3 and C22:6 ω-3, among which docosahexaenoic acid was the predominant polyunsaturated fatty acid, showing values between 4.62 and 33.11%. The presence of high concentrations of polyunsaturated fatty acids found in P. Pomum and C. brevifrons allow their recommendation for human consumption with appropriate resource utilization. Rev. Se comparó el contenido de ácidos grasos en los lípidos totales de los caracoles Phyllonotus pomum y Chicoreus brevifrons, recolectados en tres diferentes épocas del año en Punta Arena, estado Sucre, Venezuela. Las concentraciones lipídicas oscilaron entre 0.87 y 1.85%, correspondiendo el valor mínimo y el máximo a C. brevifrons colectado en lluvia y sequía, respectivamente. La esterificación de los ácidos grasos presentes en estos lípidos seguida de la cromatografía de gases permitió la caracterización y cuantificación de los ácidos grasos. En los lípidos totales de las dos especies, en todas las épocas, se observaron elevadas concentraciones de ácidos grasos insaturados (57.21-70.05%) seguido de los saturados (20.33-31.94%). Entre los ácidos grasos insaturados, predominaron los de tipo poliinsaturados, exceptuándose el extracto lipídico de P. pomum en época de transición donde las grasas monoinsaturadas fueron las mayoritarias (38.95%). Los ácidos grasos que prevalecieron fueron: C14:0, C16:0, C18:0, C20:1, C22:1 ω- 11, C22:1 ω-9, C18:3 ω-3, C20:5 ω-3 y el C22:6 ω-3, siendo este ϊltimo el predominante entre los αcidos grasos poliinsaturados, presentando porcentajes de distribuciσn comprendidos entre 4.62 y 33.11%. Debido a las altas concentraciones de ácidos grasos poliinsaturados encontradas en ambos caracoles, se recomienda su consumo para la alimentación de los seres humanos.

The alteration in the concentrations of metallic ion Pb, Zn, K, Na, Co. Fe, and Cu in the soft parts of the Biomphalaria alexandrina snails shedding Schistosoma mansoni cercariae was detected by flame atomic absorption spectrometry. Six elements Pb, Zn, K, Na, Co, and Cu were found to be present at significantly higher concentrations in cercariae-shedding snails compared with uninfected snails. The concentration of Fe ion showed non-significant decrease in the tissues of cercariae-shedding snails. Variation in the present results compared with related previous studies lead to the suggestion that the effect of trematode parasitism on fresh-water snails should not be considered universal and might be varies according to the trematode-snail combination, the organs or the tissues analyzed and the analytical method used

Using high performance liquid chromatography (HPLC) analysis it was possible to determine simultaneously the concentration of organic acids (pyruvate, lactate, succinate, fumarate, malate, acetate, propionate, acetoacetate, and ß-hydroxybutyrate) in the digestive gland and the extracellular concentration of these same acids in the hemolymph of estivating Biomphalaria glabrata, the intermediate host of Schistosoma mansoni. After a 7 day period of estivation, there was a significant increase in the tissue levels of lactate, succinate, malate and acetate compared to non-estivating snails. After 14 days of estivation, the levels of lactate and acetate were also significantly elevated. The hemolymph concentrations of pyruvate and acetate increased significantly after 7 days and acetate concentrations continued to be significantly increased up to 14 days of estivation. The other organic acids studied, such as ketone body acetoacetate and ß-hydroxybutyrate or the volatile acid propionate, did not accumulate. Their tissue concentrations, however, increased on the 7th day of estivation and reached normal levels within two weeks of estivation for some of them. One should take into consideration how the reduction in metabolism can be handled under aerobic conditions, and what role anaerobic pathways may play in both energy formation and redox balance processes

The free amino acids composition were determined in control as well as in schistosome miracidial-exposed and cercarial shedding Biomphalaria alexandrina snails using high performance liquid chromatography [HPLC]. Moreover the free amino acids profile of one-week starved snails was also investigated. Two-weeks-post exposure induced marked elevation of most of the investigated amino acids except serine, arginine and threonine. On the other hand, cerearial shedding snails induced depletion of most of the studied amino acids, accompanied by an increase in tyrosine, cystine, phenyl alanine and leucine. Starved snails show altered amino acid profile compared to age-matching control. This study revealed that amino acid profile of the Biomphalaria alexandrina snails is of critical importance for the development of Schistosoma mansoni larvae
